Biological Background: Cystatin C Function and Localization
- Cystatin C (also known as cystatin-3, gamma-trace, neuroendocrine basic polypeptide, or post-gamma-globulin) is a secreted cysteine proteinase inhibitor belonging to the type 2 cystatin superfamily.
- It functions as a local regulator of cysteine protease activity, playing a crucial role in physiological processes such as protein catabolism and antigen presentation.
- Encoded by the CST3 gene, Cystatin C is a low-molecular-weight protein (UniProt-predicted mass 15.8 kDa) that is widely distributed in body fluids, including cerebrospinal fluid, plasma, and saliva.
- Highest expression is observed in the epididymis, vas deferens, brain, thymus, and ovary, with lower levels in the submandibular gland.
- The protein is a glycoprotein and phosphoprotein, containing disulfide bonds that are essential for its structural integrity.
- Mutations in CST3 are associated with hereditary amyloidosis and age-related macular degeneration, underscoring its clinical significance.
Experimental Guidance and Technical Tips
- The immunogen comprises the full-length human Cystatin C sequence (aa 1–146), allowing recognition of multiple epitopes and potential suitability for detecting endogenous protein in WB, IF/ICC, and ELISA.
- For WB, a specific band around 13 kDa is expected; however, due to glycosylation and other modifications, the apparent molecular weight may differ from the predicted 15.8 kDa. Include positive and negative controls to confirm specificity.
- For IF/ICC, optimize fixation and permeabilization conditions, as the target is secreted; appropriate sample preparation is critical for retaining the protein.
- Validate cross-reactivity in mouse and rat tissues using appropriate tissue lysates, as cross-reactivity is reported but should be verified under your experimental conditions.
