Biological Background: CYP24A1 Function and Localization
- CYP24A1, also known as 1,25-dihydroxyvitamin D(3) 24-hydroxylase, CP24, HCAI, CYP24, HCINF1, or P450-CC24, is a mitochondrial cytochrome P450 monooxygenase encoded by the CYP24A1 gene (Gene ID: 1591).
- It plays a central role in vitamin D catabolism and calcium homeostasis by inactivating calcidiol (25-hydroxyvitamin D(3)) and calcitriol (1-alpha,25-dihydroxyvitamin D(3)) through C24- and C23-oxidation pathways. Related references: PMID:11012668 PMID:15574355 PMID:16617161
- Through the C24-oxidation pathway, it sequentially oxidizes calcitriol to the biliary metabolite calcitroic acid. Related references: PMID:15574355 PMID:24893882
- Through the C23-oxidation pathway, it converts calcidiol to 25(OH)D3-26,23-lactone. Related references: PMID:11012668 PMID:8679605
- It also preferentially hydroxylates at C-25 other vitamin D active metabolites, such as CYP11A1-derived secosteroids 20S-hydroxycholecalciferol and 20S,23-dihydroxycholecalciferol. Related references: PMID:25727742
