Biological Background: CXCR3-B Function and Localization

  • CXCR3-B is isoform 2 of the C-X-C chemokine receptor type 3 (CXCR3), also known by alternative names including G protein-coupled receptor 9 (GPR9), CKR-L2, CD183, and interferon-inducible protein 10 receptor (IP-10 receptor).
  • It functions as a receptor for the chemokine CXCL4 and mediates the inhibitory activities of CXCL9, CXCL10, and CXCL11 on human microvascular endothelial cell (HMVEC) proliferation, survival, and angiogenesis through a cAMP-mediated signaling pathway. Unlike the main CXCR3-A isoform, CXCR3-B does not promote cell chemotaxis. Related references: PMID:12782716
  • Interaction of CXCR3-B with CXCL4 or CXCL10 leads to activation of the p38 MAPK pathway, which contributes to the inhibition of angiogenesis.
  • Overexpression of CXCR3-B in renal cancer cells down-regulates expression of the anti-apoptotic protein HMOX1 and promotes apoptosis, suggesting a tumor-suppressive role.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (amino acids 1–100) of human CXCR3-B, which is unique to this isoform; thus, the antibody may distinguish CXCR3-B from other splice variants. Consider confirming specificity using isoform-specific positive and negative controls.
  • For Western blot (WB), the predicted molecular weight is 45 kDa based on the immunogen sequence; however, post-translational modifications or glycosylation may cause the endogenous protein to migrate at a different position. Validate detection conditions in the relevant sample system.
  • In immunofluorescence (IF-P), because CXCR3-B is a transmembrane receptor, permeabilization with a mild detergent (e.g., 0.1% Triton X-100) may be necessary to access intracellular epitopes; optimize fixation and permeabilization protocols.
  • For ELISA, pairing with a capture antibody targeting a different epitope may enhance sensitivity. Consider using recombinant protein standards for assay development.
  • The antibody cross-reacts with mouse CXCR3-B, but reactivity with other species has not been tested; verify cross-reactivity if using non-human/non-mouse models.
