Biological Background: CTCF Function and Localization
- CTCF (Transcriptional repressor CTCF), also designated 11-zinc finger protein and CCCTC-binding factor, is a ubiquitously expressed chromatin binding factor that is absent in primary spermatocytes.
- It binds to specific DNA sequences and regulates the three-dimensional architecture of chromatin by forming loops and anchoring DNA to the nuclear lamina, thereby organizing the genome.
Related references: PMID:16949368, PMID:18347100, PMID:18654629, PMID:19322193 - CTCF defines boundaries between active and heterochromatic DNA by binding to chromatin insulators, thus blocking inappropriate enhancer-promoter interactions.
Related references: PMID:18347100, PMID:18654629, PMID:19322193 - It plays a critical role in allele-specific expression at the imprinted IGF2/H19 gene locus, where maternal-allele binding establishes a chromatin conformation that restricts enhancer access to IGF2.
Related references: PMID:16107875, PMID:16815976, PMID:17827499 - CTCF preferentially binds unmethylated DNA, helping to maintain methylation-free zones, while its binding is prevented by CpG methylation.
Related references: PMID:18413740 - It functions as a dual transcriptional regulator, repressing MYC and BAG1, while activating the APP gene.
Related references: PMID:8649389, PMID:9591631, PMID:9407128 - CTCF is involved in sister chromatid cohesion and localizes to centromeres during mitosis, where it recruits CENPE to pericentromeric regions.
Related references: PMID:12191639, PMID:18550811, PMID:26321640 - It is considered a tumor suppressor and is essential for early embryonic development, regulating transcription in oocytes and preimplantation embryos.
Related references: PMID:12191639
Experimental Guidance and Technical Tips
- The immunogen encompasses the N-terminal region (amino acids 1–260) of human CTCF, which may enhance accessibility to the native protein in chromatin immunoprecipitation (ChIP) assays.
- For Western blotting, a predominant band corresponding to the calculated molecular weight of ~83 kDa is expected; however, post-translational modifications may result in additional bands. We recommend validating size and specificity in the relevant sample system.
- Given the reported cross-reactivity with human, mouse, and rat, users should confirm the antibody’s performance in their specific species and tissue type, as expression levels may vary.
- In immunofluorescence, CTCF exhibits nuclear localization with potential enrichment at centromeres; appropriate fixation and permeabilization conditions should be optimized for each cell line.
- When using this polyclonal antibody for IP or ELISA, consider including appropriate isotype controls and testing a range of antibody dilutions due to lot-to-lot variability.
