Biological Background: CRALBP Function and Localization

  • Retinaldehyde-binding protein 1 (CRALBP), encoded by the RLBP1 gene, is a soluble retinoid carrier essential for proper function of rod and cone photoreceptors.
  • It participates in the regeneration of active 11-cis-retinol and 11-cis-retinaldehyde from inactive 11-trans products of the rhodopsin photocycle, a process known as the visual cycle.
  • CRALBP is also involved in the de novo synthesis of these retinoids from 11-trans metabolic precursors.
  • The protein is localized to the cytoplasm and is abundantly expressed in the retinal pigment epithelium (RPE) and Mueller glial cells of the retina, as well as the pineal gland, but is not present in photoreceptor cells themselves.
  • According to UniProt, CRALBP belongs to the retinol-binding protein family and is implicated in sensory transduction and vision; it undergoes acetylation post-translationally.

Experimental Guidance and Technical Tips

  • This rabbit monoclonal antibody was raised against a synthetic peptide corresponding to amino acids 1–100 of human CRALBP (P12271). The immunogen sequence is conserved across human, mouse, and rat, consistent with the reported cross reactivity.
  • Recommended applications include Western blot (WB), immunofluorescence (IF-P), immunohistochemistry (IHC-P), and ELISA. For WB, the predicted molecular weight is 36 kDa; a band at this size should be observed in suitable lysates (e.g., retinal pigment epithelium or retina).
  • When performing IF or IHC, ensure appropriate fixation and permeabilization for cytoplasmic targets, and include relevant positive and negative controls to validate staining specificity.
  • As with any antibody, optimal working dilutions and conditions should be determined empirically by the end user in their specific assay system.
