Biological Background: TOM20 Function and Localization

  • TOM20 (Mitochondrial import receptor subunit TOM20 homolog), also known as MAS20 or MOM19, is a central receptor component of the translocase of the outer membrane of mitochondria (TOM) complex.
  • Together with TOMM22, it functions as a transit peptide receptor at the mitochondrial surface, facilitating preprotein translocation into the TOM40 pore. Related references: PMID:18331822
  • TOM20 interacts with VDAC2 and PINK1 kinase at depolarized mitochondria, stabilizing PINK1 to trigger mitophagy via PRKN recruitment. Related references: PMID:40080546
  • It is required for the translocation of cytochrome P450 monooxygenases across the mitochondrial outer membrane.
  • Subcellularly, TOM20 localizes to the mitochondrial outer membrane, consistent with its role in preprotein import.
  • The protein undergoes post-translational modifications including phosphorylation, isopeptide bond formation, and ubiquitin conjugation, which may regulate its function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 20–145 of human TOM20, covering the cytoplasmic domain; this antibody is suitable for detecting TOM20 by Western blot (WB), immunofluorescence (IF/ICC), and fluorescence-based immunohistochemistry (IF-P).
  • The antibody is directly conjugated with , enabling direct immunofluorescence detection without a secondary antibody, which may reduce non-specific binding.
  • For WB analysis, consider using mitochondrial-enriched fractions from human, mouse, or rat cell lines to enhance signal specificity.
  • Species cross-reactivity is reported for human, mouse, and rat; however, validate the antibody in your target species and sample type with appropriate positive and negative controls.
