Biological Background: Condensin Complex Subunit 1 (CNAP1) Function and Localization

  • Condensin complex subunit 1 (CNAP1), also known as CAP-D2, hCAP-D2, or XCAP-D2 homolog, is the regulatory subunit of the condensin I complex, encoded by the NCAPD2 gene in humans.
  • It is essential for mitotic chromosome condensation, converting interphase chromatin into compact mitotic chromosomes by introducing positive supercoils into DNA in the presence of topoisomerases. Related references: PMID:11136719
  • CNAP1 may target the condensin complex to DNA via its C-terminal domain, promoting resolution of DNA catenanes between sister chromatids and facilitating chromatid decatenation during anaphase.
  • The protein localizes to the nucleus and cytoplasm, and associates with chromosomes during mitosis.
  • Early in neurogenesis, CNAP1 plays a critical role in accurate mitotic chromosome condensation in neural stem cells, influencing neuron pool size and cortical development; mutations are associated with primary microcephaly. Related references: PMID:27737959
  • Post-translational modifications include phosphorylation, and the protein is subject to proteomics identification.
  • Key associated keywords: Cell cycle, Cell division, Chromosome, DNA condensation, Mitosis.

Experimental Guidance and Technical Tips

  • For Western blotting, the antibody detects a band at approximately 157 kDa; optimize blocking and antibody dilution according to standard protocols.
  • The immunogen corresponds to amino acids 100-200 of human CNAP1, a conserved region; cross-reactivity with mouse is reported, but validation in the relevant sample system is recommended.
  • For ELISA, consider direct or sandwich formats and validate the antibody's performance with appropriate positive and negative controls.
  • Ensure samples are properly prepared to avoid degradation, as the large target protein may be sensitive to proteolysis.
