Biological Background: Clusterin (CLU) Function and Localization

  • Clusterin (CLU), also known as Apolipoprotein J, Complement cytolysis inhibitor, and Testosterone-repressed prostate message 2, is a ubiquitously expressed secreted chaperone with multiple tissue-specific isoforms and alternative splice forms.
  • Functions as an ATP-independent extracellular chaperone, preventing stress-induced aggregation of blood plasma proteins and maintaining unfolded proteins in a refolding-competent state. Related references: PMID:11123922, PMID:19535339, PMID:12176985, PMID:17260971
  • Inhibits formation of amyloid fibrils by multiple proteins (APP, APOC2, B2M, CALCA, CSN3, SNCA, and LYZ variants) and facilitates receptor-mediated internalization of chaperone-client complexes for lysosomal or proteasomal degradation. Related references: PMID:12047389, PMID:17407782, PMID:17412999, PMID:21505792
  • Protects cells from complement-mediated cytolysis by inhibiting assembly of the membrane attack complex (MAC) through prevention of C9 polymerization. Related references: PMID:2780565, PMID:1903064, PMID:2601725
  • Intracellular forms interact with SCF E3 ubiquitin-protein ligase complexes to promote ubiquitination and proteasomal degradation of targets such as COMMD1 and IKBKB, thereby modulating NF-kappa-B transcriptional activity. Related references: PMID:20068069, PMID:12882985
  • A mitochondrial form suppresses BAX-dependent cytochrome c release and apoptosis by stabilizing mitochondrial membrane integrity via interaction with HSPA5. Related references: PMID:16113678, PMID:17689225, PMID:22689054
  • Plays a role in the regulation of cell proliferation (PubMed:19137541) and is involved in neuronal differentiation (via STMN3 interaction) and clearance of immune complexes during cell injury (by similarity). Related references: PMID:19137541
  • Subcellular localization encompasses secreted and intracellular compartments, including the cytoplasm, nucleus, mitochondria, endoplasmic reticulum, and secretory vesicles. Detected in blood plasma, cerebrospinal fluid, milk, seminal plasma, and colon mucosa; ubiquitously expressed with higher levels in brain, testis, ovary, liver, and pancreas.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 314–449 of human Clusterin alpha chain) targets a region that may recognize both secreted and intracellular forms of Clusterin; validate in the relevant sample system.
  • For Western blot, expect a band near 52 kDa (precursor) and under reducing conditions possibly the cleaved alpha and beta chains; consider using positive control lysates from human, mouse, or rat tissues.
  • For IHC-P, optimize antigen retrieval and blocking conditions; consider assessing Clusterin distribution in tissues such as colon, brain, or tumor samples. Cross-reactivity with mouse and rat enables validation in animal models.
  • When developing ELISA assays, pair with appropriate detection reagents and validate specificity using recombinant Clusterin protein or reference standards.
