Biological Background: CLEC2B Function and Localization
- CLEC2B, also designated Activation-induced C-type lectin (AICL), is a member of the C-type lectin superfamily and is known by alternative names CLECSF2 and IFNRG1.
- It acts as a membrane-bound ligand that specifically engages the activating receptor NKp80 (KLRF1) on natural killer (NK) cells, thereby stimulating NK-cell cytotoxicity and cytokine production.
- Physiologically, this interaction triggers the cytolysis of malignant myeloid cells that express CLEC2B, highlighting a role in immune surveillance.
- CLEC2B is predominantly expressed in lymphoid tissues and is found in most hematopoietic cell types.
- Subcellularly, the protein localizes to the cell membrane and also resides in the Golgi apparatus membrane.
- As a glycoprotein, CLEC2B features a single transmembrane helix with a signal-anchor, and its structure is stabilized by disulfide bonds.
- Post-translational modifications include ubiquitination (Ubl conjugation), though its functional significance remains to be fully characterized.
Experimental Guidance and Technical Tips
- The immunogen used (amino acids 40–120) targets the N-terminal extracellular region, which may confer specificity; validating the antibody in cell lines expressing endogenous CLEC2B is recommended.
- For Western blot (WB) applications, a band at approximately 17 kDa is expected; consider using reducing conditions and a suitable myeloid cell line as a positive control.
- Given the cross-reactivity with rat, the antibody may be applicable in rodent models; however, species-specific validation is advised.
- For ELISA, optimize coating conditions with recombinant CLEC2B protein and verify the dynamic range using a known standard.
