Biological Background: Cleaved PARP1 p25 Function and Localization
- PARP1 (Poly [ADP-ribose] polymerase 1) is a nuclear enzyme that mediates poly-ADP-ribosylation (PARylation) of target proteins, playing a central role in DNA damage repair by recognizing strand breaks and recruiting repair machinery. Related references: PMID:17177976, PMID:18055453, PMID:18172500
- Upon caspase-3/7 cleavage during apoptosis, PARP1 is cleaved into an N-terminal 25 kDa (p25) fragment and a C-terminal 89 kDa fragment; the p25 fragment promotes AIFM1-mediated apoptosis and irreversibly binds to DNA breaks, interfering with DNA repair and enhancing cell death. - PARP1 is primarily localized in the nucleus, nucleolus, and chromosomes, but can translocate to the cytoplasm during apoptosis; it contains multiple zinc-finger domains for DNA binding and an auto-poly-ADP-ribosylation domain.
