Biological Background: Gasdermin-D Function and Localization

  • Gasdermin-D (Gsdmd) is the precursor of a pore-forming protein crucial for pyroptosis, a lytic pro-inflammatory cell death. Upon cleavage by inflammatory caspases (CASP1, CASP4/CASP11) after inflammasome activation, the N-terminal fragment oligomerizes into membrane pores of 10–15 nm diameter, allowing IL-1β and IL-18 release.   • The gasdermin-D pore can also form on mitochondrial membranes, releasing mitochondrial DNA into the cytosol, and can directly kill extracellular bacteria by binding to bacterial cardiolipin and phosphoinositides. Related references: PMID:27383986, PMID:37001519
  • Beyond pyroptosis, gasdermin-D has a transcription coactivator function: in the upper small intestine, CASP3/CASP7 cleavage generates an N-terminal fragment that translocates to the nucleus, serving as a coactivator for STAT1 to induce CIITA and MHC class II, thereby maintaining food tolerance.   • Subcellular localization is dynamic: gasdermin-D is found in the cytoplasm, inflammasome complexes, plasma membrane, mitochondrion membrane, nucleus, and is secreted extracellularly after pore formation.
  • Post-translational modifications include palmitoylation, phosphorylation, ubiquitination, and glycosylation; the protein also contains isopeptide bonds and transmembrane β-strands.
  • Tissue specificity: highly expressed in mouse brain endothelial cells, linking gasdermin-D to blood-brain barrier integrity and neuroinflammatory processes.

Experimental Guidance and Technical Tips

  • Due to the immunogen design (C-terminal residues 277–376), this antibody may detect both full-length gasdermin-D (~53 kDa) and the C-terminal cleavage fragment (~22 kDa) in Western blot. Validate cleavage patterns using LPS/ATP-stimulated mouse macrophages or caspase-1/11 inhibitors.
  • For immunoprecipitation, consider using the antibody to enrich full-length gasdermin-D or its C-terminal fragment; optimize lysis and washing conditions to preserve protein interactions.
  • In ELISA applications, pairing this antibody with an N-terminal-specific detection antibody could enable discrimination between full-length and cleaved gasdermin-D.
