Biological Background: Claudin-2 Function and Localization
- Claudin-2 is a tight junction membrane protein encoded by the CLDN2 gene. It is also known as SP82.
- It functions as a paracellular channel-forming protein, polymerizing in tight junction strands to create cation- and water-selective pores, thereby regulating epithelial permeability (paracellular tight junction permeability). Related references: PMID:20460438, PMID:36008380
- In the intestinal epithelium, it facilitates sodium and water fluxes from the peritoneal side to the intestinal lumen, aiding nutrient absorption and clearing enteric pathogens as part of the mucosal immune response (By similarity).
- In the kidney, it enables passive sodium and calcium reabsorption across proximal tubules from the lumen back to the bloodstream (By similarity).
- In the hepatobiliary tract, it permits paracellular water and cation fluxes in hepatic perivenous areas and biliary epithelium, contributing to bile flow and osmotic gradient maintenance (By similarity).
- Subcellular localization: it is predominantly found at cell junctions, specifically tight junctions, and the cell membrane.
- It undergoes post-translational modifications including phosphorylation and ubiquitination, and contains disulfide and isopeptide bonds.
Experimental Guidance and Technical Tips
- For Western blot, the antibody is expected to detect a band around 25 kDa based on the immunogen design. Validate the specific band using appropriate controls in your sample system and confirm with known positive and negative samples.
- The immunogen corresponds to the C-terminal intracellular domain of human Claudin-2 (amino acids 184-230). This region is highly conserved among human, mouse, and rat, supporting cross-reactivity. When performing ELISA or WB with samples from these species, consider antigen retrieval and blocking conditions tailored to your tissue type.
- As the antibody is polyclonal, it may recognize linear and conformational epitopes. For quantitative applications like ELISA, use a standard curve with recombinant Claudin-2 protein to ensure accurate measurement.
