- In the lung, Claudin-18 is essential for alveolar fluid homeostasis, regulating alveolar epithelial tight junction composition and ion transport, and is required for lung alveolarization and maintenance of the paracellular alveolar epithelial barrier.
- Claudin-18 acts to maintain epithelial progenitor cell proliferation and organ size by regulating the subcellular localization of YAP1, restricting its nuclear accumulation and downstream target gene transcription.
- It functions as a negative regulator of RANKL-induced osteoclast differentiation, potentially via TJP2/ZO-2 relocation, and mediates the osteoprotective effects of estrogen, acting independently of RANKL signaling pathways.
