Biological Background: CDK1/2 Function and Localization
- This antibody detects CDK1 and CDK2, two closely related cyclin-dependent kinases that are central regulators of the eukaryotic cell cycle.
- CDK1 plays a key role in driving the G2-M transition by phosphorylating numerous substrates required for mitotic entry and progression. Related references: PMID:16407259 PMID:16933150 PMID:17459720
- CDK1 catalyzes lamin (LMNA, LMNB1, LMNB2) phosphorylation at mitotic onset, leading to nuclear envelope breakdown. Related references: PMID:2188730 PMID:2344612 PMID:37788673
- Its activity is tightly controlled by phosphorylation: activated by the CDK-activating kinase (CDK7/cyclin H/MAT1) and CDC25 phosphatases, while inhibited by WEE1 and PKMYT1 kinases. Related references: PMID:18480403 PMID:20360007 PMID:2188730 PMID:41100585 PMID:20395957
- CDK1 phosphorylates beta-tubulins, NEDD1, and CC2D1A to regulate microtubule dynamics, centrosome maturation, and spindle assembly during mitosis. Related references: PMID:16371510 PMID:19509060 PMID:20171170
- In apoptosis, CDK1-mediated phosphorylation of Bcl-xL promotes cell death, whereas CASP8 phosphorylation inhibits its processing and apoptosis. Related references: PMID:19917720 PMID:20937773
- CDK1 contributes to epigenetic regulation by phosphorylating EZH2, maintaining H3K27me3 marks and gene silencing. Related references: PMID:20935635
- Subcellularly, CDK1 localizes to the nucleus, cytoplasm, mitochondria, centrosome, and mitotic spindle, in line with its diverse mitotic roles.
Experimental Guidance and Technical Tips
- For Western blotting, use proliferating cell lysates (e.g., HeLa, Jurkat) as positive controls; consider cell cycle synchronization to enhance CDK1/2 signals.
- In immunohistochemistry (IHC-P), validate antigen retrieval conditions and include tissues with known proliferative indices, such as breast cancer samples.
- For immunofluorescence (IF-P), co-stain with centriolar or mitotic markers (e.g., γ-tubulin, pericentrin) to confirm localization.
- The antibody’s immunogen (CDK2 aa 199-298) is conserved in CDK1; cross-reactivity is confirmed in human and mouse samples, but validation in other species is advised.
