Biological Background: CD46 Function and Localization
- CD46, also known as membrane cofactor protein (MCP), is a type I transmembrane glycoprotein and a member of the regulators of complement activation (RCA) gene cluster.
- Serves as a cofactor for complement factor I, mediating the cleavage of C3b and C4b to protect host cells from complement-mediated damage.
- Acts as a costimulatory factor for T-cells, inducing differentiation of CD4+ T-cells into regulatory T (Treg) type 1 cells that secrete IL-10 and suppress immune responses.
- Involved in fertilization by facilitating sperm-oocyte fusion; localizes to the inner acrosomal membrane of sperm.
- Functions as a receptor for multiple pathogens, including measles virus, adenovirus subgroup B2 and Ad3, human herpesvirus 6, and pathogenic bacteria such as Neisseria and Streptococcus pyogenes (PubMed:11260136, PubMed:11971006, PubMed:7708671). - Ubiquitously expressed on all nucleated cells (absent from erythrocytes). Heavily glycosylated, contains multiple sushi domains and disulfide bonds; alternative splicing generates multiple isoforms.

Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was generated against a recombinant protein corresponding to amino acids 40-320 of human CD46, encompassing the extracellular sushi domains critical for ligand and pathogen binding. It is suitable for Western blot, IHC-P, IF/ICC, and ELISA.
- The predicted molecular weight of CD46 is approximately 44 kDa, but due to heavy glycosylation, the observed band may appear at a higher molecular weight (50–70 kDa) by Western blot. Validate the actual band size in your sample system.
- CD46 is broadly expressed in human, mouse, and rat tissues (except erythrocytes). For IHC and IF, appropriate positive controls include kidney, spleen, or placental tissues.
- Consider using this antibody to probe CD46 expression in studies of complement regulation, T-cell costimulation, or host-pathogen interactions. Verify cross-reactivity in non-human species by sequence alignment of the immunogen region.
