Biological Background: CD44 Function and Localization
- Cell-surface receptor that plays a role in cell-cell interactions, cell adhesion and migration, helping cells sense and respond to changes in the tissue microenvironment. Related references: PMID:16541107 PMID:19703720 PMID:22726066
- Engages, through its ectodomain, extracellular matrix components such as hyaluronan, collagen, growth factors, cytokines or proteases, and serves as a platform for signal transduction. Related references: PMID:18757307 PMID:23589287
- Via its cytoplasmic domain, assembles protein complexes containing receptor kinases and membrane proteases; effectors include PKN2, RAC1, RHOA, and phospholipase C, coordinating calcium mobilization and actin reorganization essential for migration and adhesion. Related references: PMID:15123640
- Upon interaction with LGALS9 ligand, activates downstream signaling components including LCK, ERK, and MAPK to promote NK cell activation. Related references: PMID:37006235
- Participates in a wide variety of cellular functions including activation, recirculation and homing of T-lymphocytes, hematopoiesis, inflammation and response to bacterial infection. Related references: PMID:7528188
- Isoform 10 (epithelial isoform) is expressed by cells of epithelium and highly expressed by carcinomas; expression is repressed in neuroblastoma cells.
- Subcellular localization includes cell membrane, cell projection microvillus, and the secreted compartment.
- Post-translational modifications: glycoprotein, proteoglycan, phosphoprotein; extensive alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 20–178 in the N-terminal extracellular domain. For Western blot, an expected band near 82 kDa may appear, but consider isoform-specific variation due to glycosylation and alternative splicing; validate under reducing and non-reducing conditions.
- For immunohistochemistry on paraffin sections, known positive controls such as human placenta or carcinoma tissues are recommended due to high endogenous CD44 expression.
- In ELISA, ensure that the coating antigen includes the 20–178 region to maintain antibody recognition.
- Pre‑incubation with recombinant CD44 ectodomain or hyaluronan can serve as a competition control to verify binding specificity in functional assays.
