Biological Background: CD43 Function and Localization
- Leukosialin (CD43, SPN) is the predominant cell surface sialoprotein of leukocytes, heavily glycosylated and expressed as multiple isoforms.
- Regulates T-cell activation and differentiation: negatively regulates Th2 differentiation and promotes IFN-gamma expression by T-cells, inducing cytokine receptor IFNGR and IL4R expression and mediating their clustering with TCR (PubMed:18036228; PubMed:24328034). - Acts as a major E-selectin ligand responsible for Th17 cell rolling on activated vasculature and recruitment during inflammation, mediating Th17-specific adhesion to E-selectin (By similarity).
- Protects cells from apoptotic signals, promoting cell survival.
- Localizes to the plasma membrane, microvilli, uropodium, and also found in the nucleus and PML bodies.
