Biological Background: Integrin beta-2 Function and Localization

  • Integrin beta-2 (ITGB2) is also known as CD18, cell surface adhesion glycoproteins LFA-1/CR3/p150,95 subunit beta, and complement receptor C3 subunit beta. It is a type I transmembrane integrin that pairs with CD11 alpha subunits (ITGAL, ITGAM, ITGAX, ITGAD) to form leukocyte-specific adhesion receptors.
  • Integrin ITGAL:ITGB2 (LFA-1) binds ICAM1, ICAM2, ICAM3, and ISG15, and is crucial for T-cell and neutrophil adhesion, immunological synapse formation, and apoptotic neutrophil clearance. Related references: PMID:1676048 PMID:23775590 PMID:38195629 PMID:29100055
  • Integrin ITGAM:ITGB2 (MAC-1) functions as a receptor for iC3b, fibrinogen, factor X, and ICAM1, facilitating complement-mediated phagocytosis, neutrophil activation, and adhesion to endothelium. Related references: PMID:1980124 PMID:21193407
  • Integrin ITGAX:ITGB2 (p150,95) binds iC3b, fibrinogen, ICAM4 on erythrocytes, and ICAM5 on neurons, contributing to erythrophagocytosis and neuro-immune interactions. Related references: PMID:16985175 PMID:10741396
  • ITGB2 is localized to the plasma membrane and membrane rafts, essential for leukocyte adhesion and signal transduction during diapedesis and immune cell trafficking.
  • Post-translational modifications include disulfide bonds, glycosylation, phosphorylation, and a pyrrolidone carboxylic acid modification; it contains EGF-like domains and requires calcium and magnesium for activity.
  • Clinically, mutations in ITGB2 cause Leukocyte Adhesion Deficiency type I (LAD-I), a primary immunodeficiency characterized by recurrent infections and impaired wound healing.
  • Tissue expression is restricted to hematopoietic cells, with high levels in neutrophils, lymphocytes, and macrophages, consistent with its role in innate and adaptive immunity. Related references: PMID:23775590 PMID:21193407 PMID:28807980

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular region (amino acids 23–125) of human CD18, which shares high homology with mouse and rat, supporting the observed cross-reactivity.
  • For WB, a band around 85 kDa is expected; consider glycosylation-related molecular weight shifts and validate in relevant leukocyte or immune cell samples.
  • IHC-P may require antigen retrieval; IF/ICC conditions should be optimized for membrane staining. ELISA detection of native or recombinant CD18 may be considered.
