Biological Background: Tyrosine-protein phosphatase non-receptor type substrate 1 (SIRPα) Function and Localization

  • Encoded by the SIRPA gene, this protein is a member of the signal-regulatory protein family and is also known as SHPS1, CD172a, BIT, MFR, MYD-1, and p84.
  • Functions as a cell surface receptor for CD47, mediating inhibitory signals that negatively regulate phagocytosis, mast cell activation, and dendritic cell maturation.
  • Also acts as a receptor for thrombospondin-1 (THBS1), stimulating SIRPA phosphorylation and NADPH oxidase-derived ROS production in non-phagocytic cells. Related references: PMID:24511121
  • Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses triggered by cell adhesion, growth factors, or insulin.
  • Plays a role in neuronal processes, including adhesion of cerebellar neurons, neurite outgrowth, and glial cell attachment, and may contribute to synaptic function.
  • Localized to the plasma membrane and is ubiquitously expressed, with highest levels in brain and on myeloid cells; lower levels in heart, placenta, lung, liver, and other tissues.
