Biological Background: Tyrosine-protein phosphatase non-receptor type substrate 1 (SIRPα) Function and Localization
- Encoded by the SIRPA gene, this protein is a member of the signal-regulatory protein family and is also known as SHPS1, CD172a, BIT, MFR, MYD-1, and p84.
- Functions as a cell surface receptor for CD47, mediating inhibitory signals that negatively regulate phagocytosis, mast cell activation, and dendritic cell maturation.
- Also acts as a receptor for thrombospondin-1 (THBS1), stimulating SIRPA phosphorylation and NADPH oxidase-derived ROS production in non-phagocytic cells. Related references: PMID:24511121
- Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses triggered by cell adhesion, growth factors, or insulin.
- Plays a role in neuronal processes, including adhesion of cerebellar neurons, neurite outgrowth, and glial cell attachment, and may contribute to synaptic function.
- Localized to the plasma membrane and is ubiquitously expressed, with highest levels in brain and on myeloid cells; lower levels in heart, placenta, lung, liver, and other tissues.
- Subject to alternative splicing and post-translational modifications including glycosylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 395–495 within the cytoplasmic domain, suggesting the antibody recognizes an intracellular epitope; permeabilization is likely required for IF/ICC detection.
- For western blot, a band near 55 kDa is expected; however, glycosylation and other modifications may cause slight shifts.
- Cross-reactivity with mouse and rat has been reported, making this antibody suitable for comparative studies across these species; validation in the specific experimental system is recommended.
- As a rabbit monoclonal antibody, it offers high affinity and lot-to-lot consistency; optimal dilutions should be determined empirically for each application.
