Biological Background: CD138/Syndecan-1 Function and Localization
- CD138, also known as Syndecan-1, is a cell surface proteoglycan that contains both heparan sulfate and chondroitin sulfate chains.
- It functions as a linker between the cytoskeleton and the interstitial matrix (By similarity).
- It regulates exosome biogenesis in concert with SDCBP and PDCD6IP (PubMed:22660413). Related references: PMID:22660413
- It is detected in placenta (PubMed:32337544) and in fibroblasts (PubMed:36213313). Related references: PMID:32337544 PMID:36213313
- Localizes to the membrane, secreted, and in extracellular exosomes.
- Post-translationally modified by addition of heparan sulfate and chondroitin sulfate; classified as a glycoprotein and proteoglycan.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 23-251), suggesting the antibody recognizes the ectodomain of CD138.
- For IHC-P, consider using heat-induced antigen retrieval in citrate or EDTA buffer; validate staining in known CD138-positive tissues (e.g., plasma cells).
- For flow cytometry, use appropriate cell permeabilization if detecting intracellular epitopes; surface staining may require live unfixed cells.
- In ELISA, the antibody may be used for capture or detection with recombinant CD138 protein; titration is recommended.
