Biological Background: Integrin alpha-M Function and Localization
- Integrin alpha-M (ITGAM), also designated CD11b, CR3A, MO1A, or MAC-1, is the alpha subunit of the leukocyte-specific integrin MAC-1 (αMβ2, CD11b/CD18). Related references: PMID:20008295, PMID:9558116
- It functions as a receptor for the iC3b fragment of complement component 3, facilitating the uptake of complement-coated particles and pathogens. Related references: PMID:20008295, PMID:9558116
- ITGAM/ITGB2 heterodimer also acts as a receptor for fibrinogen and factor X, recognizing specific peptides in the fibrinogen gamma chain.
- Regulates neutrophil migration. Related references: PMID:28807980
- In association with the beta subunit ITGB2 (CD18), ITGAM is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils. Related references: PMID:21193407
- Serves as a receptor for ICAM1, mediating adhesion between stimulated neutrophils and endothelial cells. Related references: PMID:1980124
- Predominantly expressed in monocytes and granulocytes; protein expression confirmed in neutrophils. Related references: PMID:1346576, PMID:21193407
- Localizes to the cell membrane and membrane rafts. May also play roles in phagocytosis-induced apoptosis, mast cell development, and microglial superoxide production during brain development (By similarity).
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 145–340 of CD11b/ITGAM, an extracellular region likely exposed on the native protein; consider this when designing detection strategies.
- For Western blot (WB), a band around 127 kDa is anticipated; run appropriate molecular weight markers and validate with relevant positive controls (e.g., monocyte or granulocyte lysates).
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), membrane localization is expected; optimize fixation and permeabilization conditions to preserve epitope accessibility.
- The antibody has been reported to cross-react with mouse and rat ITGAM; however, sequence homology in the immunogen region should be verified for your specific model organism.
- ELISA applications may benefit from using the recombinant immunogen or native integrin preparations as capture antigens; titrate antibody concentrations accordingly.
