Biological Background: CD11b Function and Localization
-
Integrin alpha-M (CD11b) pairs with the beta-2 subunit (ITGB2/CD18) to mediate adhesive interactions and uptake of complement-coated particles and pathogens in monocytes, macrophages, and granulocytes.
Related references: PMID:20008295, PMID:9558116 -
It functions as the receptor CR-3 for the iC3b fragment of complement component C3, likely recognizing the R-G-D peptide sequence, and also binds fibrinogen, factor X, and ICAM-1 to support leukocyte adhesion.
-
CD11b regulates neutrophil migration and, in complex with CD18, is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils.
Related references: PMID:28807980, PMID:21193407 -
Interaction with endothelial ICAM-1 promotes adhesion between stimulated neutrophils and endothelial cells.
Related references: PMID:1980124 -
In microglia, CD11b cooperates with TYROBP/DAP12 to regulate superoxide production and neuronal apoptosis during brain development.
-
Predominantly expressed in monocytes, granulocytes, and neutrophils, CD11b localizes to the cell membrane and membrane rafts.
-
The protein contains integrin-type repeats, disulfide bonds, and glycosylation sites, and undergoes alternative splicing, giving rise to multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1-100); the resulting polyclonal antibody may recognize linear epitopes, making it suitable for Western blot under denaturing conditions.
- For Western blot, the predicted molecular weight of CD11b is ~127 kDa; however, glycosylation may cause band shifts in cell lysates. Validate with appropriate positive controls.
- In ELISA applications, polyclonal antibodies can provide high sensitivity; consider pairing with a monoclonal detection antibody for sandwich ELISA development.
- Cross-reactivity with mouse and rat CD11b suggests epitope conservation; verify species-specific reactivity in your experimental system if needed.
