Biological Background: CCL21 Function and Localization

• C-C motif chemokine 21 (CCL21), also known as 6Ckine, secondary lymphoid-tissue chemokine (SLC), exodus-2, and SCYA21, is a small cytokine belonging to the CC chemokine family.
• Binds to chemokine receptors CCR7 and atypical chemokine receptor ACKR4, mediating beta-arrestin recruitment to ACKR4; does not bind CXCR3. Related references: PMID:23341447, PMID:9507024, PMID:10201891
• Exhibits chemotactic activity for thymocytes and activated T-cells, with preferential activity towards naive T-cells, but not for B-cells, macrophages, or neutrophils; also inhibits hemopoiesis.
• Predicted to be secreted and plays a role in mediating homing of lymphocytes to secondary lymphoid organs.
• Highly expressed in high endothelial venules of lymph nodes, spleen, and appendix; lower expression in small intestine, thyroid, trachea, thymus, bone marrow, liver, and pancreas.
• The mature protein has a predicted molecular mass of ~14.6 kDa, and in SDS-PAGE under reducing conditions it typically migrates as a band around 15 kDa.

Experimental Guidance and Technical Tips

• For Western blot (WB), use reducing conditions; the antibody detects a band at approximately 15 kDa corresponding to mature human CCL21. Ensure appropriate positive controls (e.g., recombinant CCL21 or lysates from lymphoid tissues).
• For immunohistochemistry (IHC-P), validate on human lymph node or spleen sections, where CCL21 is highly expressed in high endothelial venules; antigen retrieval conditions may need optimization.
• For ELISA, the antibody can be tested as a capture or detection reagent using recombinant CCL21 as standard; determine optimal working concentrations experimentally.
• Since the antibody is polyclonal and raised against a recombinant fragment (aa 24-134), it may recognize multiple epitopes of the mature secreted protein; cross-reactivity with other chemokines should be ruled out in specific assays.
