Biological Background: Cathepsin G Function and Localization
- Cathepsin G is a serine protease with trypsin- and chymotrypsin-like specificity, preferentially cleaving after Phe, Tyr, Trp, and Leu residues. Related references: PMID:29652924 PMID:8194606
- Displays antibacterial activity against both Gram-negative and Gram-positive bacteria, independent of its protease activity. Related references: PMID:2116408 PMID:2117044
- Required for recruitment and activation of platelets via the F2RL3/PAR4 platelet receptor. Related references: PMID:10702240 PMID:3390156
- Binds reversibly to B cells, CD4+ and CD8+ T cells, and NK cells, enhancing immune cell function. Related references: PMID:7842483 PMID:9000539 PMID:9536127
- Localizes to multiple subcellular compartments including cytoplasmic granules, lysosomes, cell membrane, and nucleus, reflecting its diverse functions.
- Expressed in neutrophils and B cells at the protein level. Related references: PMID:3799965 PMID:15100291
- Cleaves and activates cytokines such as IL36G and IL33, promoting chemokine expression and immune responses. Related references: PMID:30804664 PMID:22307629
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature form of human Cathepsin G (amino acids 21–255), which is expected to detect both pro- and mature forms of the protein.
- For Western blotting, a band around 29 kDa is predicted. Verify with a positive control such as neutrophil lysates.
- In immunohistochemistry (IHC-P), antigen retrieval methods may improve staining; validate the antibody in your tissue of interest.
- The antibody cross-reacts with mouse and rat Cathepsin G, but confirm cross-reactivity in your specific assay system before quantitative use.
- Since Cathepsin G is a secreted protease, consider using protease inhibitors in sample preparation to prevent degradation.
