Biological Background: CASTOR1 Function and Localization
- Encodes cytosolic arginine sensor for mTORC1 subunit 1 (CASTOR1), also known as GATS-like protein 3 (GATSL3).
- Core component of the amino acid-sensing branch of the mTORC1 signaling pathway, functioning as an intracellular arginine sensor.
- Forms homodimers or heterodimers with CASTOR2; binds and inhibits the GATOR2 subcomplex, thereby suppressing mTORC1 activity. Binding of arginine allosterically disrupts CASTOR1-GATOR2 interaction, leading to mTORC1 activation. Related references: PMID:26972053 PMID:27487210 PMID:33594058
- Localized predominantly to the cytoplasm and cytosol.
- Widely expressed across human tissues.
- Post-translational modifications include phosphorylation (Phosphoprotein) and ubiquitination (Ubl conjugation).
- Classified under keywords: 3D-structure, Cytoplasm, Phosphoprotein, Proteomics identification, Reference proteome, Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen comprises full-length human CASTOR1 (aa 1-329); for WB analysis, consider denaturing and reducing conditions to maintain epitope accessibility.
- IHC-P experiments may require antigen retrieval and tissue-specific optimization; include positive and negative controls to confirm specificity.
- In IF/ICC, confirm cytoplasmic localization; co-staining with mTORC1 pathway markers (e.g., mTOR, S6K) can provide functional context.
- ELISA protocols can be developed for CASTOR1 quantification; consider paired antibody formats for sensitive detection.
