Biological Background: Caspase-8 Function and Localization
- Caspase-8 is a key initiator caspase that triggers extrinsic apoptosis downstream of death receptors FAS/CD95 and TNFRSF1A, activating effector caspases such as CASP3 and CASP7. Related references: PMID:24813849 PMID:24813850 PMID:9654089
- Upon ligand binding, Caspase-8 is recruited via FADD to the death-inducing signaling complex (DISC), where it undergoes autoproteolytic processing to generate the active dimer. Related references: PMID:29440439
- It cleaves the pro-apoptotic BID protein, linking extrinsic apoptosis to the mitochondrial pathway by promoting cytochrome c release. Related references: PMID:9727491 PMID:9727492
- Caspase-8 acts as a critical negative regulator of necroptosis by cleaving RIPK1 at Asp-325, which suppresses its kinase activity and limits inflammation. Related references: PMID:31511692
- It can also initiate pyroptosis through cleavage of gasdermin C and D (GSDMC, GSDMD), especially following inactivation of TAK1, leading to membrane pore formation and cytokine release. Related references: PMID:30361383 PMID:30381458
- In innate immunity, Caspase-8 modulates responses by inactivating N4BP1 downstream of TLR3 and TLR4, thereby enhancing cytokine production. Related references: PMID:32971525
- Subcellularly, Caspase-8 is found in both the cytoplasm and the nucleus.
- It is widely expressed, with highest levels in spleen, thymus, lung, liver, and kidney, and lower in heart, brain, testis, and skeletal muscle.
Experimental Guidance and Technical Tips
- The immunogen spans almost full-length mouse Caspase-8 (aa 4–480), potentially recognizing both full-length and processed fragments. Validation in your relevant cell or tissue model is advised.
- For Western blot, use positive controls like mouse spleen or thymus lysates and expect a main band near 55 kDa; activation-induced cleavage products may also appear.
- In immunofluorescence, anticipate cytoplasmic and nuclear staining; include appropriate controls and validate specificity under your fixation/permeabilization conditions.
- This polyclonal antibody can be used as a detection tool in ELISA formats; consider its utility for sandwich ELISA when paired with a specific capture antibody.
- Lot-to-lot consistency should be verified, and optimal working dilutions must be determined empirically for each application.
