Biological Background: Caspase-10 Function and Localization
- Caspase-10 is a cysteine protease belonging to the caspase family, essential for the execution of apoptosis. It is expressed in most tissues, with lower levels detected in brain, kidney, prostate, testis, and colon.
- It functions as an initiator caspase in the death receptor pathway, recruited to Fas and TNFR1 receptors via the adaptor protein FADD. It may also participate in granzyme B-mediated apoptosis.
- Upon activation, caspase-10 cleaves and activates downstream effector caspases, including CASP3, CASP4, CASP6, CASP7, CASP8, and CASP9, amplifying the apoptotic signal.
- The protease hydrolyzes tetrapeptide substrates such as Tyr-Val-Ala-Asp-AMC and Asp-Glu-Val-Asp-AMC, which is typical for caspases.
- Multiple isoforms are generated by alternative splicing: isoform 7 can enhance NF-kappaB activity while inducing only slight apoptosis, and isoform C is proteolytically inactive.
- Keywords associated with caspase-10 include apoptosis, alternative splicing, hydrolase, protease, thiol protease, zymogen, and disease variant (linked to autoimmune lymphoproliferative syndrome type II).
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 220–521 of human caspase-10, covering the large catalytic domain. As a polyclonal antibody, it may recognize both pro-form (59 kDa) and processed fragments; validate band patterns with relevant controls.
- For Western blotting, consider using reducing conditions and titrate the antibody to minimize non-specific binding. Due to alternative splicing, potential isoforms may be detected.
- This antibody is suitable for ELISA; ensure proper coating conditions and use appropriate blocking to reduce background.
- Cross-reactivity is limited to human; when working with other species, perform a pilot experiment to confirm reactivity.
- Lot-to-lot consistency can be influenced by animal immunization; we recommend routine quality control with a standard lysate.
