Biological Background: CADM1 Function and Localization

• CADM1 (Cell adhesion molecule 1), also known as IgSF4, NECL-2, SynCAM, and TSLC-1, is a single-pass type I membrane protein encoded by the CADM1 gene. It belongs to the immunoglobulin superfamily and undergoes alternative splicing to generate multiple isoforms.

• In the immune system, CADM1 interacts with CRTAM on natural killer (NK) cells and CD8⁺ T-cells, promoting NK cell cytotoxicity, IFN-gamma secretion, and retention of activated CD8⁺ T-cells within lymph nodes. • In ELISA, the antibody can be considered as either a capture or detection reagent in a sandwich format; pairing with another CADM1-specific monoclonal antibody may be explored.
