Biological Background: CADM1 Function and Localization

  • CADM1 (Cell adhesion molecule 1), also known as IgSF4, NECL-2, SynCAM, and TSLC-1, is a single-pass type I membrane protein encoded by the CADM1 gene. It belongs to the immunoglobulin superfamily and undergoes alternative splicing to generate multiple isoforms.
  • Mediates Ca2+-independent homophilic cell-cell adhesion, and can also engage in heterophilic interactions with CADM3 and NECTIN3. Related references: PMID:12050160 PMID:22438059
  • In the immune system, CADM1 interacts with CRTAM on natural killer (NK) cells and CD8+ T-cells, promoting NK cell cytotoxicity, IFN-gamma secretion, and retention of activated CD8+ T-cells within lymph nodes.   • The immunogen corresponds to amino acids 45–344 of human CADM1, covering the entire extracellular domain. This design may enable recognition of both full-length and shed forms of CADM1.
  • For Western blotting (WB), a band at approximately 49–55 kDa is expected. Due to glycosylation and other post-translational modifications, the observed molecular weight may differ from the calculated 48.5 kDa. Validate the antibody with appropriate control lysates.
  • In ELISA, the antibody can be considered as either a capture or detection reagent in a sandwich format; pairing with another CADM1-specific monoclonal antibody may be explored.
  • The reported cross-reactivity with human, mouse, and rat suggests suitability for studies involving these species in settings such as tumor biology, immunology, and neuroscience. Confirm performance in the desired sample matrix.
