Biological Background: Seipin Function and Localization
- Seipin, encoded by the BSCL2 gene, is also known as Bernardinelli-Seip congenital lipodystrophy type 2 protein. It is a key regulator of lipid droplet biogenesis.
- Mediates the formation of lipid droplets during the differentiation and maturation of adipocytes. Related references: PMID:19278620, PMID:21533227, PMID:27564575
- Together with LDAF1, defines the sites of lipid droplet formation in the endoplasmic reticulum, promotes triacylglycerol nucleation, and drives budding of lipid droplets into the cytoplasm. Related references: PMID:31708432
- Regulates protein and lipid delivery from the endoplasmic reticulum into growing lipid droplets. Related references: PMID:27879284, PMID:31178403
- During early adipogenesis, BSCL2 inhibits GPAT3 and GPAT4, preventing phosphatidic acid accumulation that could antagonize PPARG and block differentiation. This inhibition is relieved by ADIG during maturation, allowing lipid droplet expansion.
- Regulates the maturation of ZFYVE1-positive nascent lipid droplets and the function of the RAB18-ZFYVE1 complex in mediating endoplasmic reticulum-lipid droplet contacts. Related references: PMID:30970241

Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (amino acids 265-363) of human BSCL2. Verify antibody specificity using appropriate controls and consider the potential for detecting isoforms or homologs.
- For WB, validate with positive lysates from human, mouse, or rat tissues known to express BSCL2 (e.g., brain, adipose tissue). The expected band is approximately 44 kDa.
- For IHC-P and IF/ICC, empirical optimization of antigen retrieval and permeabilization is recommended due to the membrane-associated localization of seipin. Use tissues with documented BSCL2 expression as positive controls.
- For ELISA, the recombinant protein immunogen may serve as a standard. Optimize coating and detection conditions to ensure sensitivity.
- As a polyclonal antibody, lot-to-lot variability may occur. Include internal reference samples and validate performance in your assay system.
