Biological Background: CD4 Function and Localization

  • CD4 is an integral membrane glycoprotein and a defining marker of T-helper cells. It is also expressed in other immune cells such as macrophages, dendritic cells, and NK cells.
  • Functions as a coreceptor for the T-cell receptor (TCR) by binding to MHC class II molecules on antigen-presenting cells, enhancing the sensitivity of TCR to peptide-MHC complexes.
  • Simultaneously interacts with TCR and MHC class II, leading to the recruitment of the Src kinase LCK to the TCR-CD3 complex, which initiates intracellular signaling cascades essential for T-cell activation, cytokine production, and differentiation.
  • Plays a critical role in thymic development of T-helper cells and drives the differentiation of monocytes into mature macrophages.
  • Contains four extracellular immunoglobulin-like domains, a single transmembrane helix, and a short cytoplasmic tail with conserved motifs for LCK binding and palmitoylation.
  • Post-translational modifications include N-linked glycosylation, disulfide bond formation, palmitoylation, and phosphorylation, which regulate its membrane localization and signaling functions.
