Biological Background: CD11b (Integrin alpha-M) Function and Localization
- Integrin alpha-M, also known as CD11b, CR3, MAC1, and CD11b/CD18, is a member of the integrin alpha chain family encoded by the Itgam gene.
- It pairs with ITGB2/CD18 to form complement receptor 3 (CR3), mediating leukocyte adhesion, phagocytosis, and migration; it binds iC3b, fibrinogen, factor X, and ICAM1.
- CD11b is predominantly expressed on the surface of monocytes, granulocytes, macrophages, and microglia, with lower expression in a subset of peritoneal mast cells.
- Subcellularly, it localizes to the plasma membrane and membrane rafts, requiring calcium and metal ions for function.
- The protein undergoes N-linked glycosylation, contains disulfide bonds, and exists in multiple isoforms due to alternative splicing.
- In the brain, CD11b/ITGAM cooperates with TYROBP/DAP12 in microglia to produce superoxide ions essential for developmental neuronal apoptosis (PubMed:18685038). Related references: PMID:18685038
Experimental Guidance and Technical Tips
- This antibody is biotin-conjugated and intended for flow cytometry; pair with streptavidin-fluorophore conjugates for detection.
- The immunogen spans amino acids 143-337 of mouse CD11b; confirm that this epitope is accessible in your experimental system.
- For optimal staining, titrate the antibody on a known positive cell line (e.g., mouse monocytes or bone marrow-derived macrophages).
- CD11b is a large glycoprotein (127 kDa); if performing intracellular staining, ensure adequate permeabilization and fixation.
- Validate cross-reactivity if used on non-mouse species, as reactivity is reported only with mouse.
