Biological Background: CD16 Function and Localization

• CD16 is a low-affinity IgG Fc receptor that exists as two isoforms: FCGR3A (CD16a), a transmembrane protein, and FCGR3B (CD16b), a GPI-anchored protein. It binds clustered antigen-IgG complexes to trigger antibody-dependent cellular cytotoxicity (ADCC) and avoids activation by free monomeric IgG.
