Biological Background: Histo-blood group ABO system transferase Function and Localization
- The protein encoded by the ABO gene is the histo-blood group ABO system transferase, also known as BGAT, GTB, NAGAT, A3GALNT, and A3GALT1. It exists in several allelic forms that determine the A, B, or O blood group phenotype by encoding glycosyltransferases with different sugar specificities.
- This enzyme catalyzes the transfer of N-acetyl-alpha-D-galactosamine (GalNAc) or galactose (Gal) in an alpha1,3 linkage to the terminal galactose of the H antigen, converting it to the A or B antigen, respectively. Individuals with the O allele lack transferase activity and express unmodified H antigen.
- It localizes to the Golgi apparatus membrane and is also secreted, consistent with its role in modifying cell surface carbohydrate antigens.
- Tissue expression is highest in testis, with additional expression in pancreas, uterus, lung, and salivary gland.
- The protein is a transmembrane glycoprotein with a signal-anchor, contains a metal-binding site for manganese, and belongs to the glycosyltransferase family.
Experimental Guidance and Technical Tips
- The immunogen for this antibody is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human BGAT. For experimental validation, consider using recombinant BGAT protein or cell lysates expressing the target protein.
- For Western blotting, the predicted molecular weight of the target is 41 kDa (as per product specifications). Optimize gel percentage and transfer conditions accordingly.
- For ELISA, ensure proper coating antigen and validate with known positive and negative human serum samples if studying blood group antigen interactions.
- Since the protein has transmembrane and secreted forms, sample preparation for WB may require consideration of the subcellular localization; Golgi-enriched fractions may enhance detection.
