Biological Background: BCL6 Function and Localization
- BCL6 is a zinc finger and BTB domain-containing transcriptional repressor (also known as BCL-5, LAZ-3, ZBTB27, ZNF51) that plays a central role in germinal center formation and antibody affinity maturation.
- It represses target genes by binding directly to the DNA sequence 5'-TTCCTAGAA-3' or indirectly by interacting with transcription factors, and recruits corepressors and histone deacetylases to mediate repression.
- In germinal center B-cells, BCL6 suppresses genes involved in differentiation, apoptosis, inflammation, and cell cycle control, enabling rapid proliferation and tolerance of DNA breaks during class switch recombination and somatic hypermutation without inducing p53-dependent apoptosis.
- BCL6 also promotes the development of follicular helper T-cells while inhibiting TH1, TH2, and TH17 differentiation, and is required for immunological memory in both T- and B-cells.

- For Western blotting, a band around 79kDa (predicted) may be observed; post-translational modifications such as acetylation or phosphorylation could result in slight mobility shifts. Consider using nuclear extracts from germinal center B-cell lines (e.g., Ramos, Daudi) as positive controls.
- In immunofluorescence/ICC, expect nuclear localization. Co-staining with B-cell markers (e.g., CD20) or germinal center markers (e.g., BCL6 itself, CD10) in lymphoid tissues may help confirm specificity.
- For ELISA applications, the antibody can be used with either the synthetic peptide or recombinant BCL6 protein as capture antigen. Titration and optimization of blocking conditions are recommended.
