Biological Background: BAZ1A Function and Localization
- BAZ1A, also known as hACF1, WCRF180, or hWALp1, serves as a regulatory subunit of the ATP-dependent ACF-1 and ACF-5 ISWI chromatin remodeling complexes. These complexes organize nucleosome arrays and slide histone octamers to regulate DNA accessibility during replication, transcription, and repair. Related references: PMID:17099699 PMID:28801535
- The ACF-1 complex has a lower ATP hydrolysis rate than ACF-5, and BAZ1A functions as a linker DNA length sensor to modulate nucleosome spacing activity. Related references: PMID:17099699 PMID:28801535

- For Western blotting, BAZ1A is a large protein (~179 kDa). Use low-percentage (e.g., 6-8%) SDS-PAGE gels and prolonged wet transfer (or semi-dry with appropriate conditions) to ensure efficient transfer.
- In ELISA, the recombinant protein immunogen can serve as a positive control for direct or indirect ELISA assays. Validate the antibody's performance in your specific sample matrix and application.
