  • ATF6 is a transcription factor precursor embedded in the endoplasmic reticulum (ER) membrane. ER stress triggers its proteolytic processing, releasing the active N-terminal fragment that translocates to the nucleus to initiate the unfolded protein response (UPR).   • For Western blot, a band near 75 kDa is expected for the precursor. Treat cells with ER stress inducers (e.g., tunicamycin, thapsigargin) to observe processing and nuclear translocation.
  • For IHC-P, consider antigen retrieval with citrate or EDTA buffer, and confirm signal specificity with adequate controls.
  • In IF/ICC, ATF6 shows a reticular ER/Golgi pattern under basal conditions and redistributes to the nucleus after stress induction; use appropriate fixation and permeabilization.
