Biological Background: ATF6 Function and Localization

  • ATF6, also known as ATF6-alpha, is a cyclic AMP-dependent transcription factor that acts as a precursor embedded in the endoplasmic reticulum (ER) membrane. Upon ER stress, it is proteolytically processed to release the active transcription factor form. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • The processed ATF6 translocates to the nucleus and initiates the unfolded protein response (UPR) by activating transcription of UPR target genes. Related references: PMID:10564271, PMID:11158310, PMID:11163209
  • ATF6 binds to the ER stress response element (ERSE) with the consensus 5'-CCAC[GA]-3' half-site, requiring NF-Y for full ERSE binding, and also binds to ERSE II. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • ATF6 may be involved in activation of transcription by the serum response factor and plays a potential role in foveal development and cone function in the retina. Related references: PMID:26029869
  • ATF6 is ubiquitously expressed in human tissues.
  • Subcellularly, the full-length ATF6 localizes to the ER and Golgi apparatus membranes, while the processed form translocates to the nucleus.
  • The protein undergoes post-translational modifications including glycosylation and ubiquitination (isopeptide bond formation).
  • Keywords: Activator, DNA-binding, Endoplasmic reticulum, Glycoprotein, Golgi apparatus, Membrane, Nucleus, Signal-anchor, Transcription, Transmembrane, Unfolded protein response.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal cytoplasmic domain (amino acids 1-194) of human ATF6, which is present in both the precursor and processed forms; consider this when designing detection assays.
  • For Western blot (WB), the predicted molecular weight of full-length ATF6 is approximately 75 kDa; validate band size in relevant sample systems under reducing conditions.
  • For immunohistochemistry (IHC-P) and immunoprecipitation (IP), antigen retrieval and antibody validation in the relevant tissue context are recommended.
  • ELISA applications may require optimized coating and detection strategies; consider testing with recombinant ATF6 fragments as controls.
