Biological Background: Histone H3 Function and Localization

Histone H3 is a core component of the nucleosome, which wraps and compacts DNA into chromatin. This structure limits DNA accessibility to the cellular machineries that require DNA as a template, thereby playing a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications (PTMs) of histones, collectively referred to as the "histone code," as well as through nucleosome remodeling.

Canonical histone H3.1, encoded by multiple genes including H3C1, H3C2, H3C3, etc., is widely expressed. In contrast, histone H3.1t (also known as H3-4 or HIST3H3) is specifically expressed in testicular cells.

At the subcellular level, histone H3 localizes primarily to the nucleus and to chromosomes.

Histone H3 undergoes extensive and varied PTMs, including acetylation, methylation, phosphorylation, citrullination, and others. Arginine residues can be methylated to form asymmetric dimethylarginine (ADMA) by protein arginine methyltransferases (PRMTs) such as PRMT6 and CARM1, which act on specific residues (e.g., R3, R18) and modulate chromatin structure and gene expression. This antibody is designed to specifically recognize asymmetric dimethylation at arginine 26 (R26) of histone H3.
