Biological Background: ASS1 Function and Localization
- Encodes argininosuccinate synthase, a key enzyme of the urea cycle that catalyzes the formation of argininosuccinate from aspartate, citrulline and ATP; together with ASL, it is responsible for arginine biosynthesis in most body tissues.
- Belongs to the urea cycle, transforming neurotoxic ammonia into non-toxic urea in the liver of ureotelic animals; plays a critical role in the ammonia detoxification pathway.
- Functionally annotated with several keywords including ATP-binding, ligase activity, and nucleotide-binding; also subject to post-translational modifications such as acetylation and phosphorylation.
- Predominantly expressed in adult liver, consistent with its role in the hepatic urea cycle; subcellularly localized to the cytoplasm/cytosol.
- Also known as citrulline--aspartate ligase (ASS; CTLN1); mutations in ASS1 are associated with citrullinemia type I, a urea cycle disorder.
