Biological Background: Aromatase (CYP19A1) Function and Localization
- Aromatase (CYP19A1) is a cytochrome P450 monooxygenase, also known as CYPXIX, Cytochrome P-450AROM, or Estrogen synthase, that catalyzes the conversion of C19 androgens (androstenedione and testosterone) to C18 estrogens (estrone and estradiol). Related references: PMID:27702664, PMID:2848247
- The enzyme carries out three successive oxidations: two conventional oxidations at C19 followed by an aromatization step that cleaves the C10-C19 bond, forming a phenolic A ring and formic acid. Related references: PMID:20385561
- Alternatively, the third oxidation can yield a 19-norsteroid and formic acid, and the enzyme also converts dihydrotestosterone to delta1,10-dehydro 19-nordihydrotestosterone, potentially contributing to androgen homeostasis. Related references: PMID:22773874

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa1-180) of Aromatase; the antibody may recognize the full-length protein and possibly some splice variants containing this region. Validate in your specific sample system.
- For Western blot, expect a band at approximately 58 kDa. Consider optimizing blocking conditions and antibody dilution for your sample type.
- For IHC-P and IF/ICC, appropriate antigen retrieval and permeabilization steps may be required.
- Cross-reactivity with rat has been observed; validate signal specificity in rat tissues if used in multi-species experiments.
- As Aromatase is localized to the endoplasmic reticulum membrane, ensure proper membrane protein extraction protocols for optimal detection.
