Biological Background: Arginase-1 Function and Localization

  • Arginase-1 (Arg-1), also known as Liver-type arginase or Type I arginase, is a key enzyme in the urea cycle that converts L-arginine to urea and L-ornithine, playing a central role in nitrogen detoxification and the biosynthesis of proline and polyamines.
  • It regulates L-arginine availability for nitric oxide synthase (NOS) in nonhepatic tissues, thereby modulating innate and adaptive immune responses, including T cell and NK cell function.
  • In group 2 innate lymphoid cells (ILC2s), Arginase-1 promotes acute type 2 inflammation and is required for optimal ILC2 proliferation (PubMed:27043409). Related references: PMID:27043409
  • In alternatively activated (M2) macrophages, it supports wound healing, tissue regeneration, defense against parasites, and immunosuppression, with organ-specific outcomes (PubMed:19360123, PubMed:20483789, PubMed:23552798). Related references: PMID:19360123, PMID:20483789, PMID:23552798
  • In tumor microenvironments, Arginase-1 expressed by dendritic cells and myeloid-derived suppressor cells (MDSCs) suppresses T cell-mediated antitumor immunity (PubMed:19414774, PubMed:23248265). Related references: PMID:19414774, PMID:23248265
  • Subcellularly, it localizes to the cytoplasm and cytoplasmic granules. It is expressed in macrophages, ILC2s, lung tumor-associated myeloid cells, and tumor-infiltrating dendritic cells.
  • As a phosphoprotein and manganese-dependent hydrolase, Arginase-1 requires metal binding for catalytic activity and is involved in both innate and adaptive immunity.

Experimental Guidance and Technical Tips

  • Based on the immunogen design (N-terminal 1-100 amino acids), this antibody may recognize the native protein in various applications.
  • For Western blot (WB), a band around 35 kDa is expected; consider using appropriate positive controls such as liver or macrophage lysates.
  • In IHC-P and IF-P, optimize antigen retrieval and permeabilization steps as the target is cytoplasmic.
  • For ELISA, validate using recombinant Arginase-1 or biological samples containing the native enzyme.
