Biological Background: AR-V7 Function and Localization
- AR-V7 is isoform 3 of the androgen receptor (AR), also known as dihydrotestosterone receptor or nuclear receptor subfamily 3 group C member 4, encoded by the AR gene (alternatively designated DHTR, NR3C4).
- The protein lacks the C-terminal ligand-binding domain and therefore constitutively regulates the transcription of a specific set of canonical AR-target genes, including PSA/KLK3 and TMPRSS2, independently of steroid hormones. Related references: PMID:19244107 PMID:25008967
- Some genes are differentially regulated by full-length AR (isoform 1) and AR-V7 (isoform 3). AR-V7-specific target genes may be regulated independently of FOXA1 expression. Related references: PMID:25008967
- AR-V7 functions as a transcription factor in the nucleus, where it binds to androgen response elements (AREs) in the absence of androgens, driving ligand-independent gene expression programs.
- At the protein level, AR-V7 is expressed in basal and stromal cells of the prostate.
- The calculated molecular weight of AR-V7 is approximately 67376 Da, consistent with the 67 kDa band observed on western blots.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a unique C-terminal sequence of AR-V7 (amino acids 544-644), which is absent in full-length AR, providing isoform selectivity. Validate specificity using appropriate AR-V7-positive and -negative cell models.
- For western blot (WB), expect a band at approximately 67 kDa. Run proper controls and consider using a loading control to confirm specificity.
- This polyclonal antibody is suitable for immunofluorescence/immunocytochemistry (IF/ICC) and ELISA applications. Optimal dilutions should be determined empirically in the relevant sample system.
