Biological Background: CD44 Function and Localization

  • CD44 is a multifunctional cell-surface receptor also known as Ly-24, Pgp-1, HERMES, ECMR-III, and GP90 lymphocyte homing/adhesion receptor. It acts as a receptor for hyaluronan (HA) and other extracellular matrix components, participating in cell-cell interactions, cell adhesion, and migration. It plays a key role in T-lymphocyte activation, recirculation, and homing, as well as in hematopoiesis, inflammation, and response to bacterial infection. Related references: PMID:25065622 PMID:8343954
  • The protein engages hyaluronan, collagen, growth factors, cytokines, and proteases through its ectodomain and assembles intracellular signaling complexes via its cytoplasmic domain, involving effectors such as PKN2, RhoGTPases RAC1 and RHOA, and phospholipase C to regulate cytoskeletal reorganization and calcium mobilization essential for cell migration.
  • CD44 is subject to extensive alternative splicing, generating multiple isoforms with distinct tissue distributions and functions.
  • It is localized primarily at the cell membrane and on microvilli, and can also be found as a secreted form.
  • Tissue expression includes the intestinal epithelium, consistent with its roles in epithelial homeostasis and immune surveillance.
  • Upon interaction with galectin-9 (LGALS9), CD44 activates downstream signaling via LCK, ERK, and MAPK pathways, promoting natural killer (NK) cell activation. Related references: PMID:37006235
  • Post-translational modifications include glycosylation, phosphorylation, and sulfation, as well as addition of glycosaminoglycan chains, classifying it as a glycoprotein and proteoglycan.
  • The protein contains disulfide bonds important for structural stability and ligand interaction.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant protein corresponding to amino acids 23-176 of the mouse CD44 ectodomain (NP_033981.2). This region includes part of the extracellular domain, likely encompassing the hyaluronan-binding link domain. Consider that the antibody may recognize an epitope within this segment.
  • This product is supplied as an APC/Cyanine7 conjugate, suitable for direct flow cytometry. Ensure appropriate instrument settings and compensation when using tandem dyes.
  • Validate antibody specificity in the relevant mouse sample system (e.g., mouse splenocytes or intestinal epithelial cells) using appropriate isotype controls and Fc receptor blocking.
  • Since CD44 is expressed on many immune cell types, analyze cell surface staining for identification or phenotyping of lymphocyte subsets.
