Biological Background: Integrin alpha-M (CD11b) Function and Localization
- Integrin alpha-M (CD11b) is the alpha subunit of the leukocyte-specific integrin ITGAM/ITGB2 (also known as CR3 or MAC-1), a receptor for complement component iC3b, as well as fibrinogen, factor X, and ICAM1.
- ITGAM/ITGB2 mediates adhesion and phagocytosis in monocytes, macrophages, and granulocytes, and regulates neutrophil migration.
- In association with ITGB2 and CD177-PRTN3, it participates in the activation of TNF-primed neutrophils.
- In microglia, ITGAM/ITGB2 is required together with TYROBP/DAP12 to produce superoxide ions that promote developmental neuronal apoptosis. Related references: PMID:18685038
- The protein is localized to the cell membrane and membrane rafts, consistent with its role in cell adhesion and signaling.
- CD11b is predominantly expressed in monocytes and granulocytes, but also found in peritoneal mast cells and microglia.

- Since the immunogen corresponds to an extracellular region (amino acids 143-337), this antibody is likely to recognize surface CD11b on live cells without permeabilization; however, validate staining conditions in your own experimental system.
- Ensure adequate compensation for the APC/Cyanine7 tandem dye, which can exhibit degradation and spillover into the APC channel under certain conditions; use fresh conjugate and include appropriate controls.
