Biological Background: IgD Function and Localization
- IgD is the constant region of delta heavy chains, existing as both membrane-bound (mIgD) and secreted (sIgD) immunoglobulins, and serves as the primary antigen receptor on peripheral B-cells, usually co-expressed with IgM. Related references: PMID:20176268, PMID:22158414
- Antigen binding to mIgD triggers B-cell clonal expansion and differentiation into plasma cells, a process involving CD79A and CD79B phosphorylation by Src family kinases. Related references: PMID:20176268, PMID:22158414
- Soluble IgD concentration in serum is lower than IgG, IgA, and IgM but higher than IgE, and its secretion is downregulated following B-cell receptor engagement.

- The APC/Cyanine7 conjugate is compatible with standard red (633–647 nm) laser lines; ensure appropriate filter sets and compensation controls are used in multicolor flow cytometry panels.
- When co-staining with IgM or other B-cell markers, verify that the fixation and permeabilization protocol does not interfere with the extracellular epitope recognized by this clone.
