Biological Background: CD4 Function and Localization

  • CD4 is an integral membrane glycoprotein also known as T-cell surface antigen T4/Leu-3, encoded by the CD4 gene in humans.
  • It functions primarily as a coreceptor for MHC class II molecules on antigen-presenting cells, interacting with the T-cell receptor (TCR) and recruiting the Src kinase LCK to initiate T-cell activation, lymphokine production, and T-helper cell differentiation.   • CD4 also plays roles in macrophages and NK cells, mediating differentiation, cytokine expression, and migration through TCR/LCK-independent pathways, and triggers monocyte-to-macrophage maturation.
