Biological Background: CD14 Function and Localization
- CD14 (monocyte differentiation antigen CD14; also known as My23 antigen or myeloid cell-specific leucine-rich glycoprotein) serves as a coreceptor for bacterial lipopolysaccharide (LPS). In concert with LBP, CD14 captures monomeric LPS and presents it to the LY96/TLR4 complex, triggering the innate immune response.
Related references: PMID:1698311, PMID:23264655, PMID:20133493, PMID:22265692 - Downstream signaling is mediated by MyD88, TIRAP, and TRAF6, culminating in NF-kappa-B activation and pro-inflammatory cytokine secretion.
Related references: PMID:8612135 - CD14 also functions as a coreceptor for TLR2 heterodimers: TLR2:TLR6 for diacylated lipopeptides and TLR2:TLR1 for triacylated lipopeptides. These interactions trigger signaling from the cell surface and subsequent targeting to the Golgi in a lipid-raft dependent pathway.
Related references: PMID:16880211 - CD14 binds electronegative LDL (LDL(-)) and mediates LDL(-)-induced cytokine release.
Related references: PMID:23880187 - CD14 localizes to the cell membrane, membrane rafts, and Golgi apparatus; a soluble form is also secreted. The protein is GPI-anchored, glycosylated, and contains characteristic leucine-rich repeats (LRRs) and disulfide bonds.
- CD14 is strongly expressed on the surface of monocytes, weakly on granulocytes, and on most tissue macrophages.

Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (aa 20–352), suggesting the antibody recognizes surface-expressed CD14; validate staining on primary monocytes or macrophage cell lines.
- For flow cytometry (FC), titrate the antibody to determine optimal staining concentration for your system; include an appropriate isotype control.
- APC/Cyanine7 offers bright far-red detection suitable for multicolor panels; ensure proper compensation and avoid fluorochrome aggregation by protecting from light.
- Cross-reactivity with human and cynomolgus monkey CD14 makes this antibody suitable for translational and non-human primate studies.
