Biological Background: TIM-3/HAVCR2 Function and Localization

  • TIM-3 (HAVCR2, CD366) is a type I transmembrane glycoprotein and immune checkpoint receptor belonging to the T-cell immunoglobulin and mucin domain (TIM) family.
  • Functions primarily as an inhibitory receptor on T-cells, suppressing Th1-mediated auto- and alloimmune responses and promoting immunological tolerance. Related references: PMID:14556005
  • Acts as a receptor for galectin-9 (LGALS9), which upon binding can induce apoptosis of antigen-specific T-cells, though this ligand-receptor interaction has been challenged. Related references: PMID:16286920, PMID:24337741, PMID:23555261
  • Also binds phosphatidylserine (PtSer) on apoptotic cells, mediating their phagocytosis by dendritic cells and modulating innate immunity. (By similarity)
  • Expressed on regulatory T (Treg) cells where it can inhibit Th17 cell responses; on natural killer (NK) cells, it has dual roles, either enhancing IFN-gamma production or suppressing cytotoxicity depending on context. Related references: PMID:24838857, PMID:22323453, PMID:22383801
  • Subcellularly located at the cell membrane and cell junctions, where it interacts with signaling molecules like LCK and ZAP70 to modulate TCR signaling. Related references: PMID:24337741, PMID:26492563
  • Tissue expression includes Th1 lymphocytes, Treg cells, dendritic cells, NK cells, and epithelial tissues; expression is upregulated in chronic viral infections such as HCV and HIV-1.
  • Post-translational modifications include glycosylation, disulfide bond formation, palmitoylation, and phosphorylation, which regulate its localization and signaling.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 22–200) of human TIM-3/HAVCR2, which targets the native receptor on the cell surface—ideal for flow cytometry applications.
  • For FC, titrate the antibody to determine the optimal staining concentration and include appropriate isotype controls to ensure specificity.
  • TIM-3 expression is typically low on resting T-cells; consider activating cells (e.g., with anti-CD3/CD28) or using samples from chronic infection/tumor models to upregulate expression for detection.
  • Validate performance in your cell system of interest; conditions such as sample preparation, fixation, and permeabilization may require optimization.
