Biological Background: CD3 Epsilon Chain Function and Localization
- CD3E encodes the CD3 epsilon chain, a subunit of the T-cell receptor (TCR)-CD3 complex, also known as T-cell surface antigen T3/Leu-4 epsilon chain.
- As part of the TCR-CD3 complex on T-lymphocytes, it plays a central role in adaptive immunity by transducing signals upon antigen recognition. Related references: PMID:15294938 PMID:15546002 PMID:2470098
- The cytoplasmic domain contains immunoreceptor tyrosine-based activation motifs (ITAMs), which become phosphorylated by LCK and FYN kinases after TCR engagement, leading to downstream signaling. Related references: PMID:2470098 PMID:40592325
- Phosphorylated ITAMs serve as docking sites for ZAP70, converting it into an active enzyme that propagates T-cell activation signals (By similarity).
- CD3E is essential for proper T-cell development and participates in internalization and down-regulation of TCR-CD3 complexes via endocytosis sequences. Related references: PMID:10384095 PMID:26507128
- Beyond its coreceptor function, CD3E acts as a receptor for ITPRIPL1, which inhibits T-cell activation by recruiting NCK1 to block ZAP70 interaction and the ERK-NFkB signaling cascade. Related references: PMID:12110186 PMID:38614099
- It contains an immunoglobulin domain, a transmembrane helix, and multiple phosphorylation sites, and is classified as a receptor with signal and disulfide bond features.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein of human CD3; the resulting monoclonal antibody is specific for the epsilon chain and validated for flow cytometry.
- For flow cytometry, consider titrating the antibody to determine optimal concentration for your cell type, and always include proper isotype controls and Fc blocking.
- Due to the APC/Cyanine7 conjugation, minimize light exposure during staining and storage to prevent photobleaching.
- Validate reactivity in your target human T-cell samples; cross-reactivity with other species has not been reported.
