Biological Background: CD69 Function and Localization
- Early activation antigen CD69 is a transmembrane receptor rapidly expressed on the surface of activated T-cells, B-cells, natural killer cells, neutrophils, platelets, and eosinophils. Related references: PMID:38540778
- It plays a central role in immune homeostasis by modulating T-cell differentiation, including negative regulation of Th17 cell development through interaction with galectin-1 on dendritic cells, which involves the JAK3/STAT5 signaling pathway and RORC transcription. Related references: PMID:20696842
- CD69 promotes regulatory T-cell conversion via the S100A8/S100A9 complex on peripheral blood mononuclear cells and acts as an oxidized low-density lipoprotein (oxLDL) receptor to induce PDCD1 expression.
- In eosinophils, it induces IL-10 production through the ERK1/2 pathway, contributing to anti-inflammatory responses. Related references: PMID:38540778
- It negatively regulates chemotactic responses by acting as a sphingosine 1-phosphate receptor 1 (S1PR1) agonist, facilitating receptor internalization and degradation. Related references: PMID:16525420
- Participates in adipose tissue-derived mesenchymal stem cell‑mediated protection against Pseudomonas aeruginosa infection by pathogen recognition and ERK1‑dependent granulocyte‑macrophage colony‑stimulating factor secretion.
- Localizes to the cell membrane as a single‑pass type II transmembrane protein and undergoes post‑translational modifications including glycosylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 64–199 of mouse CD69, covering part of the extracellular domain; this design may favor recognition of the native conformation on live cells during flow cytometry.
- For flow cytometry applications, ensure proper compensation controls are used for the APC/Cyanine7 tandem dye and that cells remain viable throughout staining and acquisition.
- CD69 is rapidly upregulated upon cellular activation; careful sample handling and timely processing are recommended to avoid unintended ex vivo activation and ensure accurate detection.
- Cross-reactivity has been confirmed for mouse; for other species, validation in the relevant biological system is advised.
